V laboratorn\u00edch podm\u00ednk\u00e1ch je izolovan\u00fd ze supernatantu tk\u00e1\u0148ov\u00e9 kultury sekre\u010dn\u00edch bun\u011bk epitelu brzl\u00edkov\u00e9ho kortexu. Prok\u00e1z\u00e1no experiment\u00e1ln\u011b m\u011b\u0159en\u00edm in vivo.<\/li>\n<li>Stimulace apopt\u00f3zy aberantn\u00edch bun\u011bk. TPA je schopen iniciovat apopt\u00f3zu v n\u011bkter\u00fdch bun\u011b\u010dn\u00fdch populac\u00edch, v\u010detn\u011b ur\u010dit\u00fdch typ\u016f krevn\u00edch bun\u011bk. I kdy\u017e je prim\u00e1rn\u011b schopen stimulovat imunitn\u00ed odpov\u011b\u010f, kter\u00e1 c\u00edl\u00ed na malign\u00ed bu\u0148ky (viz v\u00fd\u0161e), bylo experimenty in vitro zji\u0161t\u011bno, \u017ee TPA selektivn\u011b zp\u016fsob\u00ed bun\u011b\u010dnou smrt v deficitn\u00edch (infikovan\u00fdch, aberantn\u011b zm\u011bn\u011bn\u00fdch) T lymfocytech za nep\u0159\u00edtomnosti jak\u00e9koli jin\u00e9 imunitn\u00ed reakce. Je p\u0159\u00edm\u00fdm sign\u00e1lem k apopt\u00f3ze aberantn\u00edch bun\u011bk bez zapojen\u00ed dal\u0161\u00edch imunitn\u00edch proces\u016f. Tento d\u011bj je iniciov\u00e1n i v prost\u0159ed\u00ed siln\u00e9 farmakologick\u00e9 imunosuprese. Prok\u00e1z\u00e1no experimenty in vitro.<\/li>\n<li>Stimulace hematopo\u00e9zy jako kompenzace jejich deficitu a endogenn\u00ed popt\u00e1vky po krevn\u00edch bu\u0148k\u00e1ch a krevn\u00edch desti\u010dk\u00e1ch. CD4+ lymfocyty reguluj\u00ed produkci v\u0161ech typ\u016f krevn\u00edch bun\u011bk v kostn\u00ed d\u0159eni, v\u010detn\u011b \u010derven\u00fdch krvinek, krevn\u00edch desti\u010dek, granulocyt\u016f, dendritick\u00fdch bun\u011bk atd. Deficit CD4+ Th lymfocyt\u016f vede k pancytopenii pozorovan\u00e9 u subjekt\u016f s oslabenou imunitou, v\u010detn\u011b pacient\u016f s rakovinou podstupuj\u00edc\u00edch chemoterapii, u virov\u00fdch nebo jinak chemicky vyvolan\u00fdch stav\u016f. TPA je nezbytn\u00fd pro stimulaci zral\u00fdch T-lymfocyt\u016f, co\u017e vede ke zv\u00fd\u0161en\u00e9 antivirov\u00e9 nebo protin\u00e1dorov\u00e9 aktivit\u011b. Je pops\u00e1na jeho \u00fa\u010dinnost p\u0159i l\u00e9\u010db\u011b stav\u016f charakterizovan\u00fdch nedostatkem alespo\u0148 jednoho typu krevn\u00edch bun\u011bk ( \u010derven\u00fdch krvinek, krevn\u00edch desti\u010dek a granulocyt\u016f\u2026) a kompenzaci t\u011bchto stav\u016f t\u00edm, \u017ee stimuluje zv\u00fd\u0161enou tvorbu krevn\u00edch bun\u011bk pr\u00e1v\u011b typu pot\u0159ebn\u00e9ho ke zm\u00edrn\u011bn\u00ed takov\u00e9ho nedostatku. Prok\u00e1z\u00e1no experiment\u00e1ln\u011b m\u011b\u0159en\u00edm in vivo i na tis\u00edc\u00edch klinick\u00fdch p\u0159\u00edpad\u016f.<\/li>\n<\/ol>\n<p>&nbsp; &nbsp; &nbsp; T\u011bchto p\u011bt z\u00e1kladn\u00edch a nejv\u00fdznamn\u011bj\u0161\u00edch funkc\u00ed TPA v imunitn\u00edm syst\u00e9mu vysv\u011btluje pozitivn\u00ed vliv pod\u00e1v\u00e1n\u00ed TPA na pr\u016fb\u011bh n\u011bkter\u00fdch onemocn\u011bn\u00ed kauz\u00e1ln\u011b spojen\u00fdch s imunitou. Na laboratorn\u011b i klinicky prok\u00e1zan\u00fd \u00fa\u010dinek TPA navazuje v organismu ohromn\u00e1 a spletit\u00e1 kask\u00e1da modern\u00ed imunologi\u00ed popsan\u00fdch a prostudovan\u00fdch imunitn\u00edch proces\u016f a to v\u010detn\u011b v\u0161ech relevantn\u00edch zp\u011btnovazebn\u00fdch kontroln\u00edch mechanism\u016f. Th1 typ diferenciace stimulovan\u00fd TPA je krom\u011b aktivace a proliferace CD8+ T lymfocyt\u016f zodpov\u011bdn\u00fd za produkci interferonu \u03b3, TNF-\u03b2, IL-2 a IL-12 a maximalizaci \u00fa\u010dinnosti zab\u00edjen\u00ed d\u00edky aktivaci makrof\u00e1g\u016f k fagocyt\u00f3ze a \u0161t\u011bpen\u00ed intracelul\u00e1rn\u00edch bakteri\u00ed a prvok\u016f. Doch\u00e1z\u00ed k podpo\u0159e produkce IgG (opsonizuj\u00edc\u00ed protil\u00e1tky). IFN\u03b3 zvy\u0161uje produkci IL-12 dendritick\u00fdmi bu\u0148kami a makrof\u00e1gy a skrze pozitivn\u00ed zp\u011btnou vazbu, IL-12 stimuluje produkci IFN gama v Th lymfocytech a t\u00edm podporuje diferenciaci v Th1 lymfocyty. IFN\u03b3 tak\u00e9 inhibuje produkci IL-4, \u010d\u00edm\u017e inhibuje vznik Th2 lymfocyt\u016f a tak kompeti\u010dn\u011b blokuje i nadm\u011brnou aktivaci Th2 typu.<br>Pozitivn\u00ed ovlivn\u011bn\u00ed Th1 vysv\u011btluje dal\u0161\u00ed popsan\u00fd efekt, kter\u00fdm je n\u00e1sobn\u011b&nbsp;<strong>lep\u0161\u00ed a rychlej\u0161\u00ed odpov\u011b\u010f na vakcinaci<\/strong>&nbsp;proti virov\u00fdm chorob\u00e1m p\u0159i soub\u011b\u017en\u00e9m pod\u00e1n\u00ed vakc\u00edny a TPA \u2013 vznikem pam\u011b\u0165ov\u00fdch CD45RO+ Th lymfocyt\u016f.<br>Oproti um\u011bl\u00fdm Th1 shifter\u016fm, jako je nap\u0159\u00edklad inosin pranobex \u2013 ISOPRINOSIN, p\u016fsob\u00ed TPA sice podobn\u011b, ale prost\u0159ednictv\u00edm p\u0159irozen\u00fdch zp\u011btnovazebn\u011b kontrolovan\u00fdch proces\u016f a nem\u016f\u017ee tak doj\u00edt ke \u0161kodliv\u00e9mu posunu pouze sm\u011brem k Th1, ale pouze k resetu
